INSERT JARS DIRECTIONS

 

Please read carefully regarding the process/use of your Insert Jars
IF YOU HAVE A GLOVEBOX YOU CAN USE IT INSTEAD OF THE OVEN.

Inserts (Refills)

When you first get your new Insert Jars you will find that there are NO HOLES in the lids for your needle. This will insure the jars are sterile upon arrival.

1. Preheat your over to 100°f.

2. Wipe your jars down with rubbing alcohol (71% or higher) and place on a clean countertop.

3. Get 2 nails (little wider then the needle) and wipe them with alcohol and gentle tap it through the lid (ABOUT 1/4") BUT DO NOT PULL IT OUT!!!

Do this to the opposite side of the jar lid as well (now you have two nails sticking out of the jar).
You do not have to nail them all the way into the jar. Just tap the nail in about 1/4 inch or so.

4. Place the jar in the oven (leaving the oven door open) and pull out the nails.

5. Place a piece of cloth tape over the hole (First Aid isle at Wal-Mart) and take out of the oven (you do not need to leave them in the oven).

At this point the jar is no longer under pressure and will not SUCK IN air during inoculation.

You can take them out of the oven at this time.

Now when you inoculate the jars you are to pull back the tape, inject, and lay the tape right back over the hole.

Allow me to explain what this procedure is all about.

All of the jars are under pressure and will immediately suck and air once the nails are pulled out.
By preheating your oven 100°f that will push all contain nation and dust out of the oven which would leave a contaminated free area.

While the jars are in the oven and you will pulled announce out in the jar should immediately suck and air. Since the air is clean you should not have to worry about contamination being sucked in.

At that point I will cover the holes with cloth tape which will still allow air to enter but the tape will filter out most contaminants.

Now make sure you wipe down your needle with alcohol before pulling up the tape and inserting your needle. You can do this procedure on a clean to countertop or, if you have a glove box, use it.
 

Note: You will notice that your syringe has 12 cc's marked on the side of the syringe.  For every 1 cc you are to inoculate 4 holes (.25 cc) = ONE JAR.  Thus, one 12 cc spore syringe should inoculate twelve 1/2-pint jars.


Turn jar after inoculation!  Here is a little trick that I have learned recently.  Place the jars in your hands just after inoculation and quickly flick your wrist so the substrate in the jar moves a little. Try to get the substrate to 'rotate' to the other side of the jar.  That will GREATLY speed up the process due to the fact it spreads the spores throughout the jar.

Note:  Any unused spore solution can be kept in a cool place for up to 6 months.  After six months the spores can become less viable and the mycelium might not spread as quickly as one would like.

Note:  Inject ALL of the pre-sterilized substrate jars with spore syringe ASAP.  Any pre- sterilized substrate jars that have not been injected with a spore syringe is susceptible to contamination in the near future.  
Store the jars at your own risk!

Note:  Sometimes the needle will get jammed with substrate upon entry into the jar.  If this happens take the needle out and wipe it with the alcohol soaked paper towel until you see nothing on the tip of the needle and try again.
